Perfect Soul Stone for a Gargantuan Soul works slightly differently from a normal Soul stone. In order to use it (or even have it) you must be on the final step of the quest An Eternal Harvest. It must be equipped while fighting (and defeating) Giant Kralove in the Lair of the Giant Kralove to obtain the Kralove Soul quest item. The soul can then be given to Otomai to complete An Eternal Harvest and receive the Ochre Dofus.
You do not need to cast the spell Soul Capture to receive the soul. All characters with Perfect Soul Stone for a Gargantuan Soul equipped will obtain a Kralove Soul after the Giant Kralove fight.
